Pre-midterm notes

IMPORTANT ANNOUNCEMENT:

MONDAY'S 'REVIEW' (8AM) WILL BE IN ROOM 205, SCARFE BUILDING

Also: there are several very good practice questions posted on the course website!

And if you have any questions regarding recombination analysis, pedigrees, meiosis, etc, just post them in the comments. I'll check the site at least 3 times over the weekend.

Pam,

Could you please refer us to which questions on the course webpage are revelent for this exam. There are some questions when looking at the solution I could figure out but I am not sure if this the next chapter "gene interaction" or not. Therefore, learning how to do these questions would be useless. Thanks.

for the practice mt, quest.2. I am not sure if i doing it correctly. First, you find out the parental type by the two highest frequency. Then, you find which genes has crossover. You use 0.5*MII to find each gene's location to centromere.However, how can you tell if the genes are on the same side or different side of centromere since we don't know whether it is a PD or NPD? Anybody has any idea how to do this question?Thanks.

FROM THE COURSE WEBSITE THE FOLLOWING PRACTICE QUESTIONS ARE RELEVANT:

-practice midterm: question 1 (right out of your book...), question 2 (note that, although it's with a haploid, it is not a tetrad analysis question), question 3 and question 4

For the rest, you would be able to do the translocation question, but it's not relevant to your midterm (we haven't done that stuff yet)

ABOUT THE PRACTICE MIDTERM QUESTION 2

It's a haploid question, but we are given data relative to spores frequencies, not to asci frequencies. The people running the experiment opened up all the asci, took out all the spores, and found the phenotype (which is also the genotype) of each spore. Then they compiled the data table.

First off, we know that the two parental strains are different from each other. We also know that the 3 genes in question are linked. Hence, we expect 8 different classes in the offspring...see that we only have 7, and the 7th has only 1? Write out the missing class and put a "0" under the numbers column.

You are correct, the highest frequency spores are the parental ones.
You can't find MII patterns for 2 resons: 1) we are not told that the fictitious haploid fungus in question makes ordered tetrads, but most importantly 2) the data aren't presented as asci.

But back to the problem.
Now that we've got the parentals, we at least know the allelic configuration (one parent is gln-, leu+, asp- and the other is the opposite).
Next we need to find the gene order!
Remember how we do it in a 3 points testcrtoss? Here it is pretty much the same because our spores are haploid, they represent the fungus' gametes!
So, like for a 3 point testcross, look for the DCO classes...it's the class with 1 and the class with 0.
Compare them to the parentals...find the gene order.

Now you can draw out your transient diploid.

And now, I am sure you can calculate the map distances!

hi,Pam,
I am sure a lot of us bought the mt package sold by the biology society. There are a lot of polygenes and other stuff that we (or I think) didn't really cover in class.Would our mt be the same difficulty as the spring semester mt?Or should we just focus on the materials like the practice mt provided?

The biosoc package DOES NOT reflect what you are supposed to know for your MT. Except the pedigrees and the tetrad analysis stuff. The questions with the eosin eyes drosophila, translocations and inversions, modified 9:3:3:1 (15:1, 9:4:3, etc) are not applicable. In the fall they have 2 midterms and this package covers both (mainly the second one).


So, don't worry! Focus on recombination analysis (chapter 4), pedigrees, and DNA!

I didnt find the answer key for the famous gluebugs, either..Well, i thought it was a hard one and got stuck on how to insert the recombination into the perfect position.. And another question is that we only got 4 questions for midterm,rite? DNA, tetrad, 3ps test, maybe one Marker.. Do you think there would be a question on Pedigree? As we did two in midterm and besides, they are like chapter 2 material...

Yes, 4 questions.There may be a pedigree, or maybe a pedigree with markers. And for the diploid recombination stuff, not necessarily a 3 points testcross...may be a non-tescross too!

Anyway, I think you got it all covered.

it has to do with the relative position of the origin of transfer and the fertility factor genes on the plasmid...remind me to quickly go over it tomorrow!
